I've found AdAware and Spybot Search & Destroy to be very good, and they
are free. The main one I use, though, is SpySweeper by Webroot. It's a
paid service that was highly rated recently by one of the PC magazines.
I like it because it automatically downloads updates and automatically
sweeps the system like an anti-virus program.

I use Ad-Aware. Probably should add some others but it works great.
I don't get any spyware and it protects your registry amoung other
stuff.
